The CD16 and CD32b Fc-gamma receptors regulate antibody-mediated responses in mouse natural killer cells

چکیده
Abstract Natural killer (NK) cells are innate lymphocytes capable of mediating immune responses without prior sensitization. NK express Fc-gamma receptors (FcγR) that engage the Fc region IgG. Studies investigating role FcγR on mouse have been limited due to lack specific reagents. In this study, we characterize expression and biological consequences activating through their FcγR. We demonstrate most CD16 receptor, a subset also inhibitory CD32b receptor. Critically, these FcγRs functional can modulate antibody-mediated responses. characterized mice with conditional knock-out alleles Fcgr3 (CD16) or Fcgr2b (CD32b) in lymphoid cell (ILC) lineage. did not reveal any developmental defects were responsive cross-linking receptors, cytokine stimulation, killing YAC-1 targets. Physical and functional association of p56lck with Fc gamma RIIIA (CD16) in natural killer cells
The transmembrane receptor for immunoglobulin G immune complexes on natural killer (NK) cells and macrophages, Fc gamma RIIIA (CD16), mediates cellular activation through a tyrosine kinase-dependent pathway. We show that Fc gamma RIII crosslinking results in activation of the src-related kinase p56lck in NK cells and demonstrate a physical association of p56lck with Fc gamma RIIIA in immunoprec...
متن کاملIdentification of an unusual Fc gamma receptor IIIa (CD16) on natural killer cells in a patient with recurrent infections.
We found an unusual fc gamma receptor IIIa (CD16) phenotype on the natural killer (NK) cells of a 3-year-old boy, who suffered from recurrent viral respiratory tract infections since birth. He also had severe clinical problems after Bacille Calmette-Geérin (BCG) vaccination and following Epstein-Barr virus and Varicella Zoster virus infections. His peripheral blood lymphocytes contained a norma...
متن کاملSignal transduction by the CD2 antigen in T cells and natural killer cells: requirement for expression of a functional T cell receptor or binding of antibody Fc to the Fc receptor, Fc gamma RIIIA (CD16)
Crosslinking of CD2 antigen on T lymphocytes and natural killer (NK) cells leads to a rise in cytoplasmic-free Ca2+ concentration ([Ca2+]i). However, CD2 seems unlikely to interact directly with the second messenger pathways since signaling via CD2 is poor in T cells that lack the T cell receptor (TCR) and is absent in L cells or insect cells that express CD2. In contrast, NK cells that are als...
متن کاملFc gamma R(CD16) interaction with ligand induces Ca2+ mobilization and phosphoinositide turnover in human natural killer cells. Role of Ca2+ in Fc gamma R(CD16)-induced transcription and expression of lymphokine genes
In this study, we present evidence that interaction of Fc gamma R(CD16) with ligands (immune complexes or anti-CD16 antibodies) induces a rapid rise in [Ca2+]i and fast production of both inositol 1,4,5 triphosphate (IP3) and IP4 in homogeneous NK cell preparations. Part of the initial [Ca2+]i rise observed upon stimulation of NK cells with either anti-CD16 antibodies alone or after their cross...
متن کاملPositional Relationship between Natural Killer Cells and Distribution of Sympathetic Nerves in Decidualized Mouse Uterus
Background: Uterine natural killer (uNK) cells are the most abundant leukocytes in pre-implantation endometrium and early pregnancy deciduas in humans and rodents. They are associated with structural changes in maternal spiral arteries but regulation of their re-cruitment and activation is incompletely understood. The major subpopulation of uNK cells in humans expresses CD56, the neural cell ad...
